The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use 0.5µg for 105 cells. Use at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 105 to 108 cells/test.
Note: It has been observed that increased incubations times (30-45 minutes) with the CD28 antibody can enhance the staining obtained.
Involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival.
Expressed in T-cells and plasma cells, but not in less mature B-cells.
Flow cytometry staining of BALB/c splenocytes with Anti-Mouse CD3e FITC and 0.25 µg of Golden Syrian Hamster IgG Isotype Control APC (left) or 0.25 µg of ab95559 (right). Total viable cells were used for analysis.
