Our Abpromise guarantee covers the use of ab17061 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
  • Application notes
IHC-P: 1/50 - 1/100. Perform heat mediated antigen retrieval before commencing with IHC staining protocol with 10 mM Citrate buffer, pH 6.0.

    Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
  • Target

    • FunctionProbable role in assembly and expression of the TCR complex as well as signal transduction upon antigen triggering.
    • Involvement in diseaseDefects in CD247 are the cause of immunodeficiency due to defect in CD3-zeta (CD3ZID) [MIM:610163]. An immunological deficiency characterized by T-cells impaired immune response to alloantigens, tetanus toxoid and mitogens.
    • Sequence similaritiesBelongs to the CD3Z/FCER1G family.
      Contains 3 ITAM domains.
    • DomainThe ITAM domains mediate interaction with SHB.
    • Post-translational
Phosphorylated on Tyr residues after T-cell receptor triggering.
    • Cellular localizationMembrane.

    Anti-CD3 zeta antibody images

    • IHC image of ab17061 staining in Human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab17061, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
