The antibody is routinely tested by flow cytometry not by Western Blotting. The cell lines derived from B-cell chronic lymphocytic leukemia (B-CLL) should be the positive control.

You should avoid reducing conditions as you propose - the protocol is very general (classical Laemmli buffer). Use SDS gels (SDS is not reducing but denaturing agent) without adding reducing agents (bME, DTT) in the sample buffer.
