Target
-
Function
Functions as a dosage-dependent inducer in mitotic control. It is a tyrosine protein phosphatase required for progression of the cell cycle. It directly dephosphorylates CDK1 and activate its kinase activity. -
Sequence similarities
Belongs to the MPI phosphatase family.
Contains 1 rhodanese domain. -
Developmental stage
Expressed predominantly in G2 phase. -
Post-translational
modificationsPhosphorylated by CHK1 on Ser-216. This phosphorylation creates a binding site for 14-3-3 protein and inhibits the phosphatase. Phosphorylated by PLK4. -
Cellular localization
Nucleus. - Information by UniProt

All lanes : HRP Anti-Cdc25C antibody [E302] (ab205734) at 1/2500 dilution
Lane 1 :HeLa whole cell lysate (ab150035) at 10 µg
Lane 2 : Wild-type HAP1 cell lysate at 20 µg
Lane 3 : Cdc25C knockout HAP1 cell lysate at 20 µg
Developed using the ECL technique.
Performed under reducing conditions.
Predicted band size: 53 kDa
Observed band size: 60 kDa why is the actual band size different from the predicted?
Exposure time: 16 minutesThis blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab205734 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.
