  • Application notesICC/IF: 1/200 - 1/1000. Methanol-fixed cells seem to give better results than paraformaldehyde-fixed. Please see abreview for further details.
    IHC-P: 1/100. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
    WB: Use at a concentration of 1 - 5 µg/ml. Detects a band of approximately 65 kDa (predicted molecular weight: 65 kDa).

    Not yet tested in other applications.

    • FunctionInteracts with centromeric heterochromatin in chromosomes and binds to a specific subset of alphoid satellite DNA, called the CENP-B box. May organize arrays of centromere satellite DNA into a higher order structure which then directs centromere formation and kinetochore assembly in mammalian chromosomes.

Paraffin embedded human skin tissue was incubated with ab25734(1/100 dilution) for 30 mins at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.

      ab25734 was tested in a tissue microarray (TMA) containing a wide range of normal and cancer tissues. Further results for this antibody can be found at www.proteinatlas.org

    • ab25734 (1/1000) staining CENPB in HeLa cells (green). Cells were fixed with methanol and counterstained with DAPI in order to highlight the nucleus (red). Please refer to abreview for further experimental details.

    • ICC/IF image of ab25734 stained human HeLa cells. The cells were 4% PFA fixed (10 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab25734, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive IF result in HEK 293 cells.
