Applications

Our Abpromise guarantee covers the use of ab56853 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 5 µg/ml.

This antibody has only been tested in WB against the recombinant fragment used as immunogen. We have no data on the detection of endogenous protein.

Flow Cyt Use 0.1µg for 106 cells. ab170191-Mouse monoclonal IgG2a, is suitable for use as an isotype control with this antibody.

Target

  • RelevanceCEP250 (Centrosome associated protein CEP250) is a core centrosomal protein which may play an important role in centrosome cohesion during interphase. It dissociates from the centrosomes when parental centrioles separate at the beginning of mitosis. CEP250 is phosphorylated by NIMA-related kinase 2 (NEK2).
  • Cellular localizationCytoplasm, perinuclear region. Centrosome, centriole. Cell projection, cilium basal body. Note=Component of the core centrosome. In interphase cells, it specifically associates with the proximal ends of both mother and daughter centrioles. Associates with the centrosome in interphase cells. In mitotic cells, it dissociates from the mitotic spindle poles. At the end of cell division, it reaccumulates at centrosomes. In photoreceptors, found at the proximal ends of basal bodies.

  • Overlay histogram showing HepG2 cells stained with ab56853 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab56853, 0.1μg/1x106 cells) for 30 min at 22°C. The secondary antibody used was an anti-mouse Alexa Fluor® 488 (ab150113) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 0.1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.

  • Western blot against tagged recombinant protein immunogen using ab56853 CEP250 antibody at 1ug/ml. Predicted band size of immunogen is 34 kDa
