A 2 component chemiluminescent kit for the detection of horseradish peroxidase (HRP) in western blotting.
The Abcam chemiluminescent reagent for detecting proteins is extremely sensitive and has a high signal-to-noise ratio. This new non isotopic system is a high performance chemiluminescent substrate for peroxidase. It enables less than 10 picogram of protein to be detected within 30 seconds to 1 minute on exposure to X-ray film.
Instructions for use on membrane with proteins immunoblotted with peroxidase:
1) Add 1 drop of oxidant Reagent B to 1ml of substrate Reagent A (125ul of reagent covers 1cm2 membrane). This solution should be used immediately or can be kept at +4 degrees C short term.
2) Immerse the membrane in chemiluminescent reagent for 1 to 10 minutes. Remove excess reagent and cover the membrane with a plastic sheet. Put the plastic covered membrane into contact with light sensitive film in an exposure chamber. Develop the film after 30 seconds to 10 minutes exposure. The time of further radiographic exposure can be adjusted for best results and multiple copies, from the previous results.
