The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use at an assay dependent concentration.
WB Use a concentration of 0.3 - 1 µg/ml. Detects a band of approximately 23 kDa (predicted molecular weight: 26 kDa).
IHC-P Use at an assay dependent dilution. PubMed: 21454667

  • FunctionPlays a major role in tight junction-specific obliteration of the intercellular space, through calcium-independent cell-adhesion activity.
  • Tissue specificityLiver, kidney. Also found in ear.
  • Involvement in diseaseDefects in CLDN14 are the cause of deafness autosomal recessive type 29 (DFNB29) [MIM:605608]. DFNB29 is a form of sensorineural hearing loss. Sensorineural deafness results from damage to the neural receptors of the inner ear, the nerve pathways to the brain, or the area of the brain that receives sound information.
  • Sequence similaritiesBelongs to the claudin family.
  • Cellular localizationCell junction > tight junction. Cell membrane.

  • Predicted band size : 26 kDa

ab19035 (0.3 µg/ml) staining of Human Liver lysate (35µg protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.
  • ab19035 staining Claudin 14 in kidney cells by Immunocytochemistry/ Immunofluorescence.

Cells were fixed with methanol, permeabilized with Triton X-100 and blocked with 5% BSA for 30 minutes. Samples were incubated with primary antibody (1/600 in PBS + 0.5% BSA + 0.1% Triton X-100) for 1 hour at 20°C. An AlexaFluor®488-conjugated donkey anti-goat polyclonal IgG (1/800) was used as the secondary antibody.

Thank you for getting back to me. Generally if an immunogen forms an alignment of 6 or more consecutive residues I would consider it possible that the antibody would react against the homolog of the species. However, if it is a polyclonal antisera ...

Thank you for your enquiry. I have performed a sequence analysis of your fish sequence with all of immunogens used to raise our antibodies against Claudin. Unfortunately none had significant sequence similarity that would make me consider reactivit...
