IHC-P 1/200. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. In particular, boil tissue sections in 10mM citrate buffer pH 6.0 for 10 min then cool at room temperature for 20 min.

ICC/IF Use at an assay dependent concentration. PubMed: 21525338
WB 1/100. Detects a band of approximately 22 kDa (predicted molecular weight: 22 kDa).

  • FunctionPlays a major role in tight junction-specific obliteration of the intercellular space.
  • Tissue specificityExpressed in kidney, lung and prostate. Isoform 1 seems to be predominant, except in some normal prostate samples, where isoform 2 is the major form. Down-regulated in breast cancers, including ductal carcinoma in situ (DCIS), lobular carcinoma in situ (LCIS) and invasive ductal carcinoma (IDC) (at protein level), as well as in several cancer cell lines. Loss of expression correlates with histological grade, occurring predominantly in high-grade lesions.

Anti-Claudin 7 antibody images

  • Ab27487, at a dilution of 1/200, staining Claudin 7 in formalin fixed paraffin embedded human colon carcinoma tissue section by Immunohistochemistry.
  • ab27487 staining tissue sections of rat liver after oval cell activation.  The sample was PFA fixed, permeabilized in 0,1% Triton X100 for 10 minutes and blocked with 2% serum, 2%BSA, 0.05% Tween 20 in PBS for 1 hour at room temperature.  The primary antibody was diluted 1/100 and incubated for 12 hours at 4°C.  A HRP conjugated donkey anti-rabbit was used as the secondary.

  • ab27487 staining rat oval cells by ICC.  Cells were fixed in ice cold methanol for 15 minutes and blocked in 3% serum for 1 hour at 25°C prior to incubation with ab27487 (diluted 1/50 in 3% Goat serum, 2% BSA, 0.01% Tween20) for 16 hours at 4°C.  The secondary antibody was a Cy3® conjugated donkey anti-rabbit, diluted 1/100.
