ab24566 reacts with the 46 and 48 Mr subunit of the 94 Mr myelin CNPase dimer. It detects developing and adult myelin, developing oligodendrocytes and Schwann cells and distinguishes oligo endrocytes from astrocytes, microglia, neurons and other cells in brain sections.

Recommended dilutions ELISA: 1/200. IHC-P: 1/200. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. IHC-Fr: 1/200 (PMID 19799876). IHC-FoFr: 1/200. WB: 1/200. Predicted molecular weight: 47 kDa.
Note: Post-fixation in cold methanol, methanol/hydrogen peroxide or detergent is necessary for access of ab24566 to oligodendrocytes and myelin in frozen and thick Vibratome tissue sections or in tissue culture. Reaction on formalin-fixed, paraffin-embedded sections may be enhanced by autoclaving deparaffinized sections in distilled water for 10 min or by boiling sections or tissue blocks immersed in tris buffered saline, pH 9.0, in a microwave oven for 15 min.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

CNPase (2',3'-Cyclic Nucleotide 3'-Phosphodiesterase) exists in two forms: 46kDa (CNP1) and 48kDa (CNP2). It is expressed at high levels by oligodendrocytes in the central nervous system and by Schwann cells in the peripheral nervous system and is virtual absent in other cell types. CNPase activity is decreased in demyelinating diseases such as multiple sclerosis. CNPase gene is localized to chromosome 17q21, very near the BRCA 1 familial breast cancer gene.
Ab 24566 is useful for immunocytochemical studies on the progression of normal and pathologic myelination.
