The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
1/50 - 1/100. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
FunctionRequired for centriole duplication. Inhibition of centriole duplication leading to defects in cytokinesis.
Tissue specificityWidely expressed (at protein level). Highly expressed in testis. Also expressed in spleen, thymus, prostate, small intestine, colon and peripheral blood leukocytes.
Developmental stagePreferentially incorporated into the newly assembled daughter centriole during centriole assembly at the late G1 or early S phase. Remains in the daughter centrioles throughout the cell cycle. At the next cycle of centriole duplication, its amount on the original daughter centriole eventually decreases.
Post-translational modificationsPhosphorylated upon DNA damage, probably by ATM or ATR.
Cellular localizationCytoplasm > cytoskeleton > centrosome > centriole. Centriole-associated, asymmetrically localizes to the daughter centriole.
