    Collagen Type VI from adult human and bovine tissues

  • General notesAt least 11 genetically distinct gene products are collectively referred to as 'collagen types' or other proteins and proteoglycans of the extracellular matrix. In humans, collagens are composed of about 20 unique protein chains which under go various types of post-translational modifications and are ultimately assembled into a triple helix. This results in great diversity between collagen types. Collagens are highly conserved throughout evolution and are characterized by an uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. For these reasons it is often extremely difficult to generate antibodies with specificities to collagens. The development of type specific antibodies is dependent on NON-DENATURED three-dimensional epitopes. This preparation results in a native conformation of the protein.

  • Involvement in diseaseDefects in COL6A1 are a cause of Bethlem myopathy (BM) [MIM:158810]. BM is a rare autosomal dominant proximal myopathy characterized by early childhood onset (complete penetrance by the age of 5) and joint contractures most frequently affecting the elbows and ankles.
    Defects in COL6A1 are a cause of Ullrich congenital muscular dystrophy (UCMD) [MIM:254090]; also known as Ullrich scleroatonic muscular dystrophy. UCMD is an autosomal recessive congenital myopathy characterized by muscle weakness and multiple joint contractures, generally noted at birth or early infancy. The clinical course is more severe than in Bethlem myopathy.

Anti-Collagen VI antibody (Biotin) images

  • ab6583 staining Collagen VI (red) in Bovine bone marrow stromal cells (BMSCs) and articular chondrocytes (ACs) by ICC/IF (Immunocytochemistry/immunofluorescence). Sections were fixed with formalin and blocked with 5% BSA for 30 minutes. Samples were incubated with primary antibody (10 μg/ml). An Alexa Fluor®-conjugated anti-rabbit IgG was used as the secondary antibody. Decorin - green, DNA - blue.

  • ab6583 staining Collagen VI in rabbit decalcified collagen defective femur by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).
    Tissue was fixed in formaldehyde and an enzymatic antigen retrieval step was performed using Proteinase K. Samples were then blocked using 100% avidin/biotin for 10 minutes and then incubated with ab6583 at a 1/50 dilution for 1 hour. Streptavidin-HRP was used for detection of biotinylated primary.
