Application Abreviews Notes
WB Use a concentration of 0.3 - 1 µg/ml. Detects a band of approximately 60 kDa (predicted molecular weight: 53 kDa). Approx 60kDa band observed in MOLT4 and Jurkat lysates. In transfected HEK293 a band of ~50kDa is observed.
ICC/IF Use at an assay dependent concentration. See Abreview.

Target

  • FunctionMay be a crucial component of the cytoskeleton of highly motile cells, functioning both in the invagination of large pieces of plasma membrane, as well as in forming protrusions of the plasma membrane involved in cell locomotion. In mycobacteria-infected cells, its retention on the phagosomal membrane prevents fusion between phagosomes and lysosomes.

  • Cellular localizationCytoplasm > cytoskeleton. Cytoplasm > cell cortex. Cytoplasmic vesicle > phagosome membrane. In non-infected macrophages, associated with the cortical microtubule network. In mycobacteria-infected macrophages, becomes progressively relocalized and retained around the mycobacterial phagosomes. Retention on the phagosomal membrane is strictly dependent on mycobacterial viability and not due to impaired acidification.

  • ab14787 staining Coronin 1a in human glioblastoma cell line D54MG by Immunocytochemistry/ Immunofluorescence.
    Cells were fixed in paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 0.5% BSA for 20 minutes at room temperature, then incubated with ab14787 at a 1/50 dilution for 16 hours at 4°C. The secondary used was a TRITC conjugated rabbit anti-goat polyclonal, used at a 1/50 dilution. Nuclei are counterstained with DAPI.
