We recently purchased ab15191 to identify COX-2 in cat eyes. Based on sequence homology, should this antibody work? Thus far, there is little convincing positing staining in adult feline eyes or kidneys. Thanks for your help!

The lab comparedthe immunogen sequence for ab15191 with that of the cat COX2 sequence provided and the percentage homology is 41%. It is therefore very unlikely that the antibody will cross- react.

I contacted the lab to get more information regarding the immunogen,but unfortunately it is propriety. However, if you could send us the cat's protein sequence, they are happy to find out the homology for you.

"Unfortunately the immunogen sequence for ab15191 is confidential, it is located within aa 514-604 of P35355 (Uniprot). I could not locate the sequence for the cat COX2 protein on Uniprot, if the customer is willing to provide the sequence I would be happy to calculate the percentage homology with the immunogen."

The specific immunogen sequence is confidential in this case, however, I can confirm it is located within amion acids 514-604 of rat COX2, from the protein databasepage P35355 (Uniprot).

Extraband around 64 kDa in Western blot with mouse skin samples, IHC seems to show correct staining pattern.
