Application Abreviews Notes
WB 1/2000. Detects a band of approximately 46 kDa (predicted molecular weight: 46 kDa).
ELISA Use at an assay dependent concentration.
IHC-P 1/2000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • RelevanceCreatine Kinase MB consists of a dimer of nonidentical chains. With MM being the major form in skeletal muscle and myocardium, MB existing in myocardium, and BB existing in many tissues, especially brain. Creatine Kinase MB reversibly catalyses the transfer of phosphate between ATP and various phosphogens. The creatine kinase isoenzymes play a central role in energy transduction in tissues with large fluctuating energy demands such as skeletal muscle, heart, brain and spermatozoa.

Anti-Creatine Kinase MB antibody [46A1] images

  • IHC image of Creatine Kinase MB  staining in human heart muscle formalin fixed paraffin embedded tissue section, performed on a Leica Bond system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab72002, 1/200 dilution, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
