Applications

Our Abpromise guarantee covers the use of ab121653 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P 1/50 - 1/200. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol. Use buffer at pH 6 for antigen retrieval.
ICC/IF Use a concentration of 1 - 4 µg/ml. Fixation/Permeabilization: PFA/Triton X-100

Target

  • FunctionMajor structural component of the ciliary rootlet, a cytoskeletal-like structure in ciliated cells which originates from the basal body at the proximal end of a cilium and extends proximally toward the cell nucleus. Contributes to centrosome cohesion before mitosis.
  • Sequence similaritiesBelongs to the rootletin family.
  • Post-translational
    modifications
Phosphorylated by NEK2 which may regulate its association with centrosomes. Phosphorylated upon DNA damage, probably by ATM or ATR.
  • Cellular localizationCytoplasm > cytoskeleton > centrosome > centriole. In ciliated cells, associated with ciliary rootlets. In non-ciliated cells, localized between, around and at the proximal ends of the centrioles. Dissociates from the centrioles at the onset of mitosis and reassociates with them at anaphase.
