The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1 - 5 µg/ml. This antibody has only been tested in WB against the recombinant fragment used as immunogen. We have no data on the detection of endogenous protein.
Use at an assay dependent concentration.
FunctionShows moderate inhibition of cathepsin B but is not active against cathepsin C.
Tissue specificityRestricted to the stratum granulosum of normal skin, the stratum granulosum/spinosum of psoriatic skin, and the secretory coils of eccrine sweat glands. Low expression levels are found in the nasal cavity.
Sequence similaritiesBelongs to the cystatin family.
Post-translational modificationsSubstrate for transglutaminases. Acts as an acyl acceptor but not as an acyl donor.

ab54572 staining CST6 in Human hypertrophic prostate tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 4% serum for 15 minutes at 20°C; antigen retrieval was by heat mediation with citrate buffer (10mM pH6). Samples were incubated with primary antibody (1/100 in 1%BSA + PBS) for 16 hours at 20°C. An undiluted Biotin-conjugated Horse universal polyclonal was used as the secondary antibody.
