Properties

Applications

Our Abpromise guarantee covers the use of ab54728 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 5 µg/ml. This antibody has only been tested in WB against the recombinant fragment used as immunogen. We have no data on the detection of endogenous protein.
IHC-P Use a concentration of 3 µg/ml.
ICC/IF Use at an assay dependent concentration. PubMed: 20801503

Target

  • Function
May affect the rate of fibrils formation.
  • Involvement in disease
Defects in DCN are the cause of congenital stromal corneal dystrophy (CSCD) [MIM:610048]. Corneal dystrophies are inherited, bilateral, primary alterations of the cornea that are not associated with prior inflammation or secondary to systemic disease. Most show autosomal dominant inheritance.
  • Sequence similarities
Belongs to the small leucine-rich proteoglycan (SLRP) family. SLRP class I subfamily.
    Contains 12 LRR (leucine-rich) repeats.
  • Post-translational
    modifications
The attached glycosaminoglycan chain can be either chondroitin sulfate or dermatan sulfate depending upon the tissue of origin.
  • Cellular localization
Secreted > extracellular space > extracellular matrix.

Images

  • ab54728 staining Decorin in Human lung tissue sections from chronic obstructive pulmonary disease (COPD) patients by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Panel G represents micrographs from the central airways (bronchi) and panel H shows the small airways and parenchyma. L denotes lumen of the bronchi, and A and V denote airway and vessels, respectively. Black arrowheads show staining in the lamina reticularis. Scale bars represent 100 μm.

Tissue was fixed with 4% paraformaldehyde and blocked with 2% BSA-TBS for 30 minutes. Samples were incubated with primary antibody and a Biotin-conjugated Horse anti-mouse polyclonal was used as the secondary antibody. Nuclei was stained blue with hematoxylin

  • Western blot against tagged recombinant protein immunogen using ab54728 Decorin antibody at 1ug/ml. Predicted band size of immunogen is 65 kDa
  • Decorin antibody (ab54728) used in immunohistochemistry at 3ug/ml on formalin fixed and paraffin embedded human adrenal gland.
