    DNMT1 (DNA (cytosine-5)-methyltransferase 1) belongs to the DNMT family, responsible for methylation of CpG islands in the genome. DNMT1 preferably methylates cytosines located in hemimethylated DNA and it associates with DNA replication sites in S-phase.

     DNMT1 Assay Kit (ab113469) allows the user to quickly and consistently measure the amount of DNMT1. The kit is ready-to-use and provides all the essential components needed to carry out a successful DNMT1 assay experiment without the need for electrophoresis and transfer process. ab113469 is suitable for measuring DNMT amounts quantitatively from fresh tissue and cultured cells of human and mouse.

    • Nuclear extracts were prepared from MCF-7 cells using the Nuclear Extraction Kit (ab113474) and DNMT1 activity was measured using ab113469. DNMT3A and DNMT3B activities were measured using ab113470 DNMT3A Assay Kit and ab113471 EpiSeeker DNMT3B Assay Kit.

    We used the DNMT1 assay kit (ab113469) to quantify DNMT1 in nuclear fractions of H9c2 cardiomyoblasts. The objective was to compare the amount of DNMT1 in doxorubicin-treated cells with the controls. The cells were cultivated in DMEM medium and drugged with 0,025 µM of doxorubicin for 24 hours. To have a considerable signal we needed to use 6 micrograms of nuclear protein enriched fraction. The kit was easy to use and the results were consistent.
    As negative aspects of this kit we point out the fact that the assay instructions are incomplete and some reagents are not even mentioned in the procedure.
