Anti-Dopamine beta Hydroxylase antibody images
Ab43868 at 1/10000 dilution staining rat tissue; positive neurons in rat brainstem, vibratome sectioned, ABC / DAB detection.
ICC/IF image of ab43868 stained PC12 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab43868, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

ab43868 staining Dopamine beta Hydroxylase in 12 week old murine ovary tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).
Tissue was fixed with Modified Davidson's Fixative and a heat mediated antigen retrieval step was performed using sodium citrate. Samples were then permeabilized using 0.1% Tween in TBS, blocked with 10% serum for 1 hour at 23°C and then incubated with ab43868 at a 1/200 dilution for 12 hours at 4°C. The secondary used was a biotinylated goat anti-rabbit IgG (H+L) used at a 1/200 dilution.
ab43868 was used to detect Dopamine beta Hydroxylase in rat brain tissue for Electron Microscopy (EM). The Tissue was Perfusion fixed with 4% PFA and the stored for less than 1 week in 1% PFA refrigerated, tissue blocked with PBS-BSA for 15 min, incubation was 1 hour with primary antibody at room temperature. A 10nm Gold-conjugated antirabbite polyclonal(1/20) was used a secondary antibody.
