General notesStorage in frost-free freezers is not recommended.
If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
Working dilution samples should be discarded if not used within 12 hours.
Storage instructionsShipped at 4°C. Store at +4°C short term (1-2 weeks). Upon delivery aliquot. Store at -20°C or -80°C. Avoid freeze / thaw cycle.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application notesGSA: Use at an assay dependent dilution.
IP (denatured): Use at an assay dependent dilution.
WB: Use at a concentration of 0.5 - 1 µ g/ml. This concentration is determined using whole extract of transfected 293T (human embryonal kidney) cells expressing E2F4. Detects a band of approximately 50 kDa (predicted molecular weight: 44 kDa) (for natural molecule, may vary in a recombinant preparation).
Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
FunctionTranscription activator that binds DNA cooperatively with DP proteins through the E2 recognition site, 5'-TTTC[CG]CGC-3' found in the promoter region of a number of genes whose products are involved in cell cycle regulation or in DNA replication. The DRTF1/E2F complex functions in the control of cell-cycle progression from G1 to S phase. E2F-4 binds with high affinity to RBL1 and RBL2. In some instances, can also bind RB protein.
Tissue specificityFound in all tissue examined including heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas.
Sequence similaritiesBelongs to the E2F/DP family.
Developmental stagePresent in the growth-arrested state, its abundance does not change significantly as cells move into and through the cell cycle.
Post-translational modificationsDifferentially phosphorylated in vivo.
