Can be used in studies with NIH3T3 whole cell lysate, + / - PDGF stimulation (pair), PC12 whole cell lysate, + / - Sorbitol treatment and blocked using ERK 1 + 2 phospho T185 / 202 + Y187 / 204 but not non-phospho peptides (pair)
    Flow Cyt: Use 10µl for 106 cells.
    IHC-Fr: Use at an assay dependent dilution.

      Peptide Competition: Extracts prepared from PC12 cells stimulated with NGF were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF. Membranes were blocked with a 5% BSA-TBST buffer overnight at 4oC, then were incubated with 0.50 µg/mL ab4819 antibody for two hours at room temperature, following prior incubation with: no peptide (1), the non-phosphopeptide corresponding to the immunogen (2), a generic phosphothreonine containing peptide (3), a generic phosphotyrosine containing peptide (4), or, the phosphopeptide immunogen (5). After washing, membranes were incubated with goat F(ab’)2 anti-rabbit IgG alkaline phosphatase and bands were detected using the Tropix WesternStar method. The data show that only the peptide corresponding to ab4819 blocks the antibody signal, thereby demonstrating the specificity of the antibody.
