  • FunctionBinds to an ERR-alpha response element (ERRE) containing a single consensus half-site, 5'-TNAAGGTCA-3'. Can bind to the medium-chain acyl coenzyme A dehydrogenase (MCAD) response element NRRE-1 and may act as an important regulator of MCAD promoter. Binds to the C1 region of the lactoferrin gene promoter. Requires dimerization and the coactivator, PGC-1A, for full activity. The ERRalpha/PGC1alpha complex is a regulator of energy metabolism.

    Phosphorylation on Ser-19 enhances sumoylation on Lys-14 increasing repression of transcriptional activity.
    Sumoylated by SUMO2. Main site is Lys-14 which is enhanced by phosphorylation on Ser-19, cofactor activation, and by interaction with PIAS4. Sumoylation enhances repression of transcriptional activiy, but has no effect on subcellular location nor on DNA binding.

Anti-Estrogen Related Receptor alpha antibody - ChIP Grade images

  • Detection of Estrogen Related Receptor alpha binding at ESRRA and ACADM promoters by ab16363 in ChIP. MCF7 breast epithelial adenocarcinoma cells were treated with estrogen or vehicle, followed by chromatin extraction and crosslinking. The ChIP was performed using ab16363 at a concentration of 0.26 µg/µg chromatin. The DNA was quantified using Real-time PCR. Please see abreview by Genpathway Inc for additional details.

  • Estrogen Related Receptor alpha was immunoprecipitated using 0.5mg MCF7 whole cell extract, 5µg of Rabbit polyclonal to Estrogen Related Receptor alpha and 50µl of protein G magnetic beads (+). No antibody was added to the control (-).
    The antibody was incubated under agitation with Protein G beads for 10min, MCF7 whole cell extract lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.
    Proteins were eluted by addition of 40µl SDS loading buffer and incubated for 10min at 70oC; 10µl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab16363.
    Secondary: Mouse monoclonal [SB62a] Secondary Antibody to Anti-Rabbit HRP (IgG light chain) (ab99697).
    Band: 48kDa: Estrogen Related Receptor alpha.

  • ab16363 staining Estrogen Related Receptor alpha in mouse heart tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with paraformaldehyde and a heat mediated antigen retrieval step was performed using TRIS/EDTA pH 9.0. Samples were then blocked with 2% normal goat serum for 45 minutes at 22°C followed by incubation with the primary antibody at a 1/100 dilution for 16 hours at 4°C. A biotin-conjugated rabbit polyclonal was used as secondary antibody at a 1/200 dilution.
