WB Use a concentration of 0.05 - 1 µg/ml. Predicted molecular weight: 62 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.

  • FunctionTyrosine phosphatase that specifically dephosphorylates 'Tyr-142' of histone H2AX (H2AXY142ph). 'Tyr-142' phosphorylation of histone H2AX plays a central role in DNA repair and acts as a mark that distinguishes between apoptotic and repair responses to genotoxic stress. Promotes efficient DNA repair by dephosphorylating H2AX, promoting the recruitment of DNA repair complexes containing MDC1. Its function as histone phosphatase probably explains its role in transcription regulation during organogenesis. Coactivates SIX1, and seems to coactivate SIX2, SIX4 and SIX5. The repression of precursor cell proliferation in myoblasts by SIX1 is switched to activation through recruitment of EYA3 to the SIX1-DACH1 complex and seems to be dependent on EYA3 phosphatase activity (By similarity). May be involved in development of the eye.

Anti-EYA3 antibody images

  • ICC/IF image of ab22835 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab22835, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • Anti-EYA3 antibody (ab22835) at 0.1 µg/ml + HepG2 cell lysate with 5% skim milk in PBS

    HRP conjugated anti-rabbit IgG at 1/50000 dilution

    Predicted band size : 62 kDa
    Observed band size : 44 kDa (why is the actual band size different from the predicted?)
    12% gel concentration.
  • Immunohistochemistry detection of EYA3 in paraffin embedded human skeletal muscle tissue section. ab22835 was used at 4 ug/ml. Arrows indicate positively labelled muscle cells.
