Target

  • RelevanceMembers of the F-box protein family, such as Fbp5B, are characterized by an approximately 40-amino acid F-box motif. SCF complexes, formed by SKP1 (MIM 601434), cullin (see CUL1; MIM 603134), and F-box proteins, act as protein-ubiquitin ligases. F-box proteins interact with SKP1 through the F box, and they interact with ubiquitination targets through other protein interaction domains (Jin et al., 2004 [PubMed 15520277]).

    In panels A and B, there are two identical membranes (with an in
    vitro translated Fbp5b (Fbp5b ivt) and a whole cell extract from 293T cells loaded in duplicate) without  (A) and with (B) peptide blocking.

    Peptide inhibition completely removes the in vitro translated band and very strongly reduces the endogenous band from 293T cells.

    In a repeat of this experiment a mock in vitro translation was included and no band was seen in that lane.
