    Filaggrin is initially synthesized as a large, insoluble, highly phosphorylated precursor containing many tandem copies of 324 AA, which are not separated by large linker sequences. During terminal differentiation it is dephosphorylated and proteolytically cleaved. The N-terminal of the mature protein is heterogeneous, and is blocked by the formation of pyroglutamate.
    Undergoes deimination of some arginine residues (citrullination).

Anti-Filaggrin antibody [SPM181] images

  • ab17808 staining Filaggrin in Human skin tissue sections by Immunohistochemistry (IHC-P - paraffin-embedded sections). Tissue was fixed with 10% NBF and blocked with 0.025% serum free protein block for 5 minutes at 25°C; antigen retrieval was by heat mediation with a TRIS/EDTA buffer (pH9). Samples were incubated with primary antibody (1/50 in serum free antibody diluent) for 30 minutes at 25°C. An undiluted HRP-conjugated Goat polymer was used as the secondary antibody.

  • Overlay histogram showing A431 cells stained with ab17808 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab17808, 1µg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879x) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in A431 cells fixed with 100% methanol (5 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.

Tissue was fixed with formaldehyde; antigen retrieval was by heat mediation with a TRIS/EDTA buffer (pH8.2). Samples were incubated with primary antibody (1/200 in antibody diluent) for 32 minutes at 37°C. A rabbit anti-mouse monoclonal (1/500) was used as the secondary antibody.

Thank you for contacting Abcam regarding ab17808 and ab80820. I can confirm that ab17808 has not been tested to react with profilaggrin so we cannot be sure if it will cross react. On the other hand, we expect ab80820 to react with both forms. ...
