Applications

Our Abpromise guarantee covers the use of ab13493 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 3 - 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
ICC/IF Use at an assay dependent concentration. PubMed: 18363778
WB Use a concentration of 0.3 - 1 µg/ml. Detects a band of approximately 48 kDa (predicted molecular weight: 49 kDa).Can be blocked with Human Flotillin 1 peptide (ab23215).

Target

  • FunctionMay act as a scaffolding protein within caveolar membranes, functionally participating in formation of caveolae or caveolae-like vesicles.
  • Sequence similaritiesBelongs to the band 7/mec-2 family. Flotillin subfamily.
  • Cellular localizationCell membrane. Membrane > caveola. Melanosome. Endosome. Membrane-associated protein of caveolae. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

Anti-Flotillin 1 antibody images

  • ICC/IF image of ab13493 stained Malme-3M cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA, 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells. The cells were then incubated with the antibody ab13493 at 5µg/ml overnight at +4°C. The secondary antibody (green) was DyLight® 488 donkey anti- goat (ab96931) IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Product Wall

A BLAST search of the immunogen used to generate ab13493 revealed no homology of this sequence with flotillin 2 therefore I would not expect the antibody to cross react with flotillin 2.
