Flow Cyt Use 1µg for 106 cells. ab170190-Mouse monoclonal IgG1, is suitable for use as an isotype control with this antibody.
IHC-FoFr 1/500 - 1/5000. According to the materials and methods by Carlsson et al. (2005) Brain 128:559-569 "the midbrain was fixed in 4% paraformaldehyde (PFA), in 0.1 M phosphate buffer, pH 7.4, for 24h and dehydrated in 25% sucrose and cut on a freezing microtome" therefore fixation was by immersion, not perfusion.
WB Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 46 kDa (predicted molecular weight: 40.6 kDa).

  • ab11959 Fos B antibody (1/1000) immunostaining for Fos B in rat brain cortex. Staining was also detected in the striatum as expected (not shown). Rats were treated with high dose amphetamine (5mg/kg) 30 min prior to transcardial perfusion to induce Fos B expression. The staining procedure was perfomed according to the TH staining in Carlsson et al. (2005) Brain 128:559-569 with the exception that the quenching step was excluded from the staining.

  • Overlay histogram showing HeLa cells stained with ab11959 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab11959, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.
