  • FunctionProbable transcription factor. Plays a critical role in the control of immune response.
  • Involvement in diseaseDefects in FOXP3 are the cause of immunodeficiency polyendocrinopathy, enteropathy, X-linked syndrome (IPEX) [MIM:304790]; also known as X-linked autoimmunity-immunodeficiency syndrome. IPEX is characterized by neonatal onset insulin-dependent diabetes mellitus, infections, secretory diarrhea, trombocytopenia, anemia and eczema. It is usually lethal in infancy.

    Lane 1 : Jurkat Tag cells transfected with the expression vector
    Lane 2 : Jurkat Tag cells transfected with the FOXP3 expression construct


    Predicted band size : 47 kDa
    Observed band size : 47 kDa
    Additional bands at : 100 kDa. We are unsure as to the identity of these extra bands.
    24 hours after transfection, cells were lysed and analyzed by Western blot using ab54501
  • ab54501 staining FOXP3 in Mouse thymus tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 1% BSA for 20 minutes at 25°C; antigen retrieval was by heat mediation in a citrate buffer. Samples were incubated with primary antibody (1/3000 in 1% BSA/PBS) for 1 hour at 25°C. An undiluted HRP-conjugated Goat polyclonal was used as the secondary antibody.

  • ab54501 staining FOXP3 in mouse spleen tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with methanol and blocked with 5% serum for 30 minutes at 25°C. Samples were incubated with primary antibody (1/500 in 5% serum) for 24 hours at 4°C. ab150064, a donkey anti-rabbit Alexa Fluor® 594 (IgG H&L, 1/1000) was used as the secondary antibody.
