Description

Mouse monoclonal [100/3] to gamma Adaptin

Immunogen

Full length protein (Cow).

Reacts with

Hu, Cow

Does not react with
(species key)

Ms, Rat

Specificity

The antibody reacts with polypeptides of approximately 100 kDa in bovine liver, human heart fibroblasts, and Madin-Darby bovine kidney cultured cells (MDBK), but not with any components in the 110-115 kDa range from these sources, or from rat pheochromocytoma cultured cells (PC12), neuroblastoma or astrocytes.
This suggests that the 110 and 115 kDa polypeptides may be specific variants that occur only in some cell types of brain.
The antibody does not recognize the gamma-subunit in rat and mouse.
Using immunofluorescent staining on MDBK cells, and African green monkey kidney cells the binding of the antibody appears to be largely confined to the trans-Golgi network.

Relevance

Adaptins are important components of clathrin-coated vesicles transporting ligand-receptor complexes from the plasma membrane or from the trans-Golgi network to lysosomes. The adaptin family of proteins is composed of four classes of molecules named alpha, beta, beta prime and gamma adaptins. Adaptins, together with medium and small subunits, form a heterotetrameric complex called an adaptor, whose role is to promote the formation of clathrin-coated pits and vesicles. The protein encoded by this gene is a gamma-adaptin protein and it belongs to the adaptor complexes large subunits family.
