Description

Rabbit polyclonal to GFAP - Astrocyte Marker

Specificity

This antibody labels the astrocytes in the brain. In peripheral neural system it reacts with Shwann cells, satellite cells, and enteric glial cells. It is positive with astrocytomas in the central neural system. The antibody has been solid-phase absorbed with human and cow serum proteins. In crossed immunoelectrophoresis using 50 µL antibody per cm2 gel area, no reaction with 2 µL human plasma and 2 µL cow serum is observed. The antibody shows one distinct precipitate (GFAP) with cow brain extract. In indirect ELISA, the antibody shows no reaction with human plasma and cow serum.

GFAP, a class-III intermediate filament, is a cell-specific marker that, during the development of the central nervous system, distinguishes astrocytes from other glial cells.

Tissue specificity

Expressed in cells lacking fibronectin.

Involvement in disease

Defects in GFAP are a cause of Alexander disease (ALEXD) [MIM:203450]. Alexander disease is a rare disorder of the central nervous system. It is a progressive leukoencephalopathy whose hallmark is the widespread accumulation of Rosenthal fibers which are cytoplasmic inclusions in astrocytes. The most common form affects infants and young children, and is characterized by progressive failure of central myelination, usually leading to death usually within the first decade. Infants with Alexander disease develop a leukoencephalopathy with macrocephaly, seizures, and psychomotor retardation. Patients with juvenile or adult forms typically experience ataxia, bulbar signs and spasticity, and a more slowly progressive course.

ab7779 at a dilution of 1/1000, staining GFAP in astrocytes (Alexa 488 secondary at 1/2000) on 30µm coronal rat brain tissue sections in free floating IHC (see protocol link for detailed description). NB: No labeling observed following omission of primary antibody.

Sections were viewed using an Axioplan 2 Imaging microscope (Imaging Associates) fitted with 10x, 20x and 40x Plan-Neofluorobjectives (Zeiss, Germany) and images were taken using a AxioCam Hrm digital camera (Zeiss, Germany) and AxioVision software (Imaging Associates).

ab7779 staining human brain tissue by ICC/IF.  The section was formaldehyde fixed and blocked with 3% serum (for 1 hour at 24°C) prior to incubation with ab7779 (1/1000) for 12 hours at 24°C.  An Alexa Fluor® 546 conjugated goat anti-rabbit was used as the secondary antibody.
