Properties

Applications

Our Abpromise guarantee covers the use of ab95250 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use a concentration of 5 µg/ml.
WB Use a concentration of 1 µg/ml. Detects a band of approximately 41 kDa (predicted molecular weight: 41 kDa).
IHC-FoFr Use a concentration of 1 - 5 µg/ml.

Target

  • Function
Receptor for ghrelin, coupled to G-alpha-11 proteins. Stimulates growth hormone secretion. Binds also other growth hormone releasing peptides (GHRP) (e.g. Met-enkephalin and GHRP-6) as well as non-peptide, low molecular weight secretagogues (e.g. L-692,429, MK-0677, adenosine).
  • Tissue specificity
    Pituitary and hypothalamus.
  • Involvement in disease
Defects in GHSR may be a cause of idiopathic short stature autosomal (ISSA) [MIM:604271]. Short stature is defined by a subnormal rate of growth.
  • Sequence similarities
Belongs to the G-protein coupled receptor 1 family.
  • Cellular localization
    Cell membrane.

Anti-Ghrelin Receptor antibody images

  • ICC/IF image of ab95250 stained SKNSH cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab95250, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • Anti-Ghrelin Receptor antibody (ab95250) at 1 µg/ml + Human brain tissue lysate - total protein (ab29466) at 10 µg

    Secondary
Goat Anti-Rabbit IgG H&L (HRP) preadsorbed (ab97080) at 1/5000 dilution
    Developed using the ECL technique

Antibody ab92520 is raised against a ghrelin receptor amino acid sequence that is found in both isoforms of the human receptor, so it should be capable of recognizing both. Antibody ab85104 is raised against a sequence derived from rat receptor that is...
