This antibody reacts with the mu chain of human IgM. It does not react with other human Igs. It is specific for human IgM when tested against human IgA, IgG, IgM, Bence Jones kappa and lambda myeloma proteins. Electrophoresis of the antibody preparation followed by diffusion versus anti-goat IgG and anti-goat whole serum results in single arcs of precipitation.
The antibody was isolated from goat anti-human IgM antiserum by immunospecific purification to remove essentially all goat serum proteins, including immunoglobulins, which do not specifically bind to the m-chain of human IgM.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Dot: A minimum dilution of 1:6,000 was determined by direct dot blot assay using 40 ng human IgM/dot. A minimum dilution of 1:100,000 was determined in a direct chemiluminescence assay using 20 ng human IgM/dot. Luminol plus enhancer was used as substrate.
ELISA (direct): 1/50000.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
