Ihave have received another replyfrom the supplying lab, with a few more details for question 2) especially:

#1) Detection polymer composition is proprietary and that detail cannot be disclosed.

#2) We have not run our polymer detection side-by-side with a non-polymer HRP detection to allow us to give a quantitative answer. We can say that the polymer detection affords a significant increase in sensitivity that will allow users to increase primary antibody titers and/or decrease incubation times.

Unfortunately, the answer to the first question (number of HRP molecules per polymer) is proprietary.

As for the secondquestion (% increase in signal intensity compared to regular HRP-secondary antibody): The answer would be subject tomany varying conditions, such asfixation, signal in tissue, section of tissue, HIER, etc. We do not have a general answer that applies to these circumstances in this case.

I have already purchased the secondary Ab ab2891 - now I realise it is pre-diluted for IHC. If I want to use it for Western Blotting what dilution can I use it at?

Thank you for your enquiry.

Unfortunately since it has not been previously tested in Western blot and since we do not know the concentration of the secondary antibody (this information is considered confidential by the antibody originator), I can only make a guess at the dilution you would need to use.

You may wish to try a few different dilution, I might try a range from 1/10 to 1/100 to begin with, and then see what the staining looks like and adjust the dilutions accordingly.

Please do let us know how you make out with this antibody in Western blot, as we would love to know whether it will work in this application and what dilution can be used.

Can you tell me the concetration of this antibody? Can you tell me what the protein carrier and preservative are? If all of this information is proprietary, can you at least tell me if the protein carrier or preservative are likely to cross-react with galectin-3?

This antibody is a polymer detection for mouse and rabbit antibodies on human tissue. It will cross-react with virtually all monoclonals and rabbits antibodies. Specific cross-reactivity with a particular protein has not been observed. As I mentioned on the telephone, the concentration, protein carrier and preservative are proprietary.
