The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 35 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

ab171870 - Rabbit polyclonal IgG, is suitable for use as an isotype control with this antibody.

  • FunctionReceptor for short chain fatty acids through a G(i)-protein-mediated inhibition of adenylyl cyclase and elevation of intracellular calcium. The rank order of potency for agonists of this receptor is acetate= propionate = butyrate > pentanoate = formate.
  • Tissue specificityExpressed at relatively high levels in peripheral blood leukocytes and, to lesser, extent, in spleen.
  • Sequence similaritiesBelongs to the G-protein coupled receptor 1 family.
  • Cellular localizationCell membrane.

  • Human Lung, Alveolar Macrophages (formalin-fixed, paraffin-embedded) stained with ab118449 at 35 µg/ml followed by biotinylated goat anti-rabbit IgG secondary antibody, alkaline phosphatase-streptavidin and chromogen.
  • ab118449 staining GPCR GPR43 in 4µm parasagittal FFPE sections from adult mouse brain and spleen tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Antigen retrieval was by heat mediation, pH6.0. Tissue was blocked with 5% BSA for 1 hour at 20°C. Samples were incubated with primary antibody (1/200 in 1M PBS + 5% BSA) for 24 hours at 4°C. An Alexa Fluor® 647-conjugated donkey anti-rabbit IgG polyclonal (1/500, 2 hours at 20°C) was used as the secondary antibody. Second sections were incubated with the anti-Iba-1 antibody for 24 h (dilution 1/500 at 4 °C), followed by the Alexa Fluor® 568-conjugated secondary antibody staining (1/500 for 2 hours at 20°C). Nuclei were counterstained with 4,6-diamidino-2-phenylindole (DAPI).
