This antibody gave a positive signal in HepG2 and Caco2 whole cell lysates.
IF/ICC: HCT116 cell line.

If you would like information about the formulation of a specific lot, please contact our scientific support team who will be happy to help.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1 µg/ml. Detects a band of approximately 33 kDa (predicted molecular weight: 35 kDa).
Use a concentration of 10 µg/ml.
FunctionReceptor for bile acid. Bile acid-binding induces its internalization, activation of extracellular signal-regulated kinase and intracellular cAMP production. May be involved in the suppression of macrophage functions by bile acids.
Tissue specificityUbiquitously expressed. Expressed at higher level in spleen and placenta. Expressed at lower level in other tissues. In digestive tissues, it is expressed in stomach, duodenum, ileocecum, ileum, jejunum, ascending colon, transverse colon, descending colon, cecum and liver, but not in esophagus and rectum.
Sequence similaritiesBelongs to the G-protein coupled receptor 1 family.
ICC/IF image of ab114081 stained HCT116 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab114081, 10µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
Western blot - Anti-GPCR TGR5 antibody (ab114081)
All lanes : Anti-GPCR TGR5 antibody (ab114081) at 1 µg/ml
Exposure time : 2 minutesThe predicted molecular weight of GPCR TGR5 is 35 kDa (SwissProt), however we expect to observe a banding pattern at 33 kDa. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
