Description

Mouse monoclonal [BA-G5] to heavy chain cardiac Myosin

Specificity

Although this antibody was raised against myosin from skeletal muscle in cows, it has been shown to be specific for heavy chain cardiac myosin in other species (see references). It has been reported previously that BA-G5 is specific for the alpha isoform of heavy chain cardiac myosin, however other reports suggest that it also reacts with the beta isoform (PubmedID: 10412087).

Relevance

Cardiac MHC exists as two isoforms in humans, alpha-cardiac MHC and beta-cardiac MHC. These two isoforms are expressed in different amounts in the human heart. During normal physiology, beta-cardiac MHC is the predominant form, with the alpha-isoform contributing around only 7% of the total MHC. Mutations of the MHC genes are associated with several different dilated and hypertrophic cardiomyopathies.

ab50967 staining mouse heart tissue sections by IHC-P.  Sections were formaldehyde fixed and subjected to heat mediated antigen retrieval in citrate buffer pH 6, prior to blocking with sequential peroxidase and protein block (prediluted) for 20 minutes at 20°C.  The primary antibody was diluted 1/100 and incubated with the sample for 45 minutes at 20°C.  A HRP conjugated goat anti-mouse antibody was used as the secondary.

ab50967 staining heavy chain cardiac Myosin in murine HL-1 cells (derived from an atrial tumor) by Immunocytochemistry/ Immunofluorescence.

Cells were fixed in paraformaldehyde, permeabilized with 0.5% Triton X-100, blocked with 10% serum for 30 minutes at 21°C and then incubated with ab50967 at a 1/200 dilution for 16 hours at 4°C. The secondary used was a goat anti-mouse polyclonal conjugated to Alexa-Fluor 488, used at a 1/400 dilution. Counterstained with DAPI.
