  • Epitope
    This antibody recognises an epitope within aa20-30. Proline 26 is not essential for recognition by the antibody. See Gorka et al. 1998 for more information.
  • Positive control
    • Murine erythroleukemia (MEL) cells were used in part of the original characterisation of this antibody. Purified histones can also be used as a positive control. This antibody also gave a positive signal in human breast adenocarcinoma tissue sections. This antibody gave a positive signal in HeLa (Triton-enriched) cell lysate.

    Histones H1 are necessary for the condensation of nucleosome chains into higher-order structures. The H1F0 histones are found in cells that are in terminal stages of differentiation or that have low rates of cell division.

    Exposure time: 10 seconds
  • IHC image of Histone H1.0 staining in a formalin fixed, paraffin embedded human breast adenocarcinama tissue section*, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab11079, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

     

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

  • Western blot using ab11079 on HeLa whole cell extract (30 µg/lane).

    Exposure 5 sec using ECL.
    12.5% SDS-PAGE gel.

  • Mouse monoclonal to Histone H1.0 against recombinant human H1 isoforms H1.0 to H1.5 as depicted in the figure.

    Dilution 1/1000.
