Application Abreviews Notes
IHC-P Use a concentration of 1 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
ChIP Use at an assay dependent concentration.
WB 1/2000 - 1/10000. Detects a band of approximately 19 kDa (predicted molecular weight: 11 kDa).
IP Use at 1-4 µg/mg of lysate.

Target

Anti-Histone H4 antibody - ChIP Grade images

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human breast carcinoma tissue labelling Histone H4 with ab70701 at 1/200 (1µg/ml). Detection: DAB.
  • IHC image of ab70701 staining Histone H4 in human colon formalin fixed paraffin embedded tissue sections, performed on a Leica Bond. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab70701, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. No primary antibody was used in the negative control (shown on the inset).

     

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times

  • All lanes : Anti-Histone H4 antibody - ChIP Grade (ab70701) at 1/2000 dilution

    Lane 1 : Histone extract from HeLa cells at 50 µg/ml
    Lane 2 : Histone extract from HeLa cells at 15 µg/ml
    Lane 3 : Histone extract from HeLa cells at 5 µg/ml


    Predicted band size : 11 kDa
    Observed band size : 19 kDa (why is the actual band size different from the predicted?)
