Recommended dilutions ICC/IF: Use at an assay dependent concentration. Previous customer feedback suggested that this antibody did not work in ICC/IF in interphase HeLa and SK-N-SH cells as no specific binding was detected(see Abreivews). More recent testing gave a positive result with this antibody. This difference may be due to different batches being tested. We welcome any further feedback about this antibody
WB: Use at a concentration of 2 µg/ml. Detects a band of approximately 26 kDa (predicted molecular weight: 22 kDa).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

HP1 alpha is a component of heterochromatin. It recognizes and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. HP1 alpha may interact with lamin B receptor (LBR). This interaction can contribute to the association of the heterochromatin with the inner nuclear membrane. HP1 proteins are relatively small proteins (~25 kDa) with a conserved amino-terminal chromo domain and a structurally related carboxy-terminal motif, the chromo shadow domain. Both domains of HP1 are required for binding to native chromatin in vivo, but they contribute differentially to binding in euchromatin and heterochromatin.

Immunocytochemistry/ Immunofluorescence - HP1 alpha antibody (ab16482)
ICC/IF image of ab16482 stained human HeLa cells. The cells were 4% PFA fixed (10 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab16482, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal donkey serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 donkey anti-goat IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
