Applications

Our Abpromise guarantee covers the use of ab8326 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ELISA Use at an assay dependent concentration.
ICC Use at an assay dependent concentration.
WB Use a concentration of 1 µg/ml.
Functional Studies Use at an assay dependent concentration. Suitable for preparation of peroxidase-antiperoxidase detection systems - PAP complexes.

Target

  • Relevance
Horseradish Peroxidase (HRP) is an enzyme commonly used as an indicator for chemical reactions which produce peroxide. The enzyme is routinely conjugated to antibodies for use in enzyme-based immunoassay systems. HRP functions in the removal of H2O2 (hydrogen peroxide), oxidation of toxic reductants, biosynthesis and degradation of lignin, suberization, auxin catabolism, response to environmental stresses such as wounding, pathogen attack and oxidative stress. These functions might be dependent on each isozyme/isoform in each plant tissue.
  • Cellular localization
Secreted Probable. Vacuole Probable. Note: Carboxy-terminal extension appears to target the protein to vacuoles.

  • Anti-HRP antibody [HP-03] (ab8326) at 1 µg/ml + Purified horse radish peroxidase sample

    Secondary
    Goat anti-mouse IgG (H+L)-HRP conjugate.

    Performed under non-reducing conditions.
  • HeLa cells were transfected with control or ADF + cofilin1 siRNA (control data shown) followed by transfection with ss-HRP–Flag. HRP was accumulated in the trans-Golgi network (TGN) at 20°C for 2 hour in the presence of 100 µM cycloheximide. The cells were shifted to 32°C for the indicated time, and the location of HRP was detected with ab8326 (red). Cells were also stained with an anti-TGN46 (green) antibody to visualize the TGN. Bars, 5 µm.

All the information we have for this antibody is on the datasheet. The only tip we have to pass on is that as the HRP will be expressed intracellularly, a permeabilisation step will be necessary. If you are having trouble with the antibody, let me know...

I suspect that you are using a chromagen which is alcohol soluble, for instance AEC. In this case you need to use an aqueous mounting media and not an organic one such as DPX. Do not dehydrate and clear the sections, instead mount them straight from wa...

We regret to say that we can find no specific information for a suggested dilution on frozen/paraffin embedded tissues. I would personally try it at 1:50 and adjust it accordingly from the results obtained. If you have any problems with the immunohisto...

The antibody was raised against HRP, not to a peptide sequence, no studies have been performed on the antigenic determinant. The antibody can be used for the preparation of PAP complexes so it should not inhibit the activity of HRP.
