  • General notes

    Tissue: Colon Location: Rectum Diagnosis: Adenocarcinoma, well differentiated Stage: II T3NoMo Grade: I Gross findings: Tumor size 4 x 3 x 2 cm. Cut section is soft, white. Regional lymph nodes: 3. Microscopic findings: Tissue sections show proliferation of malignant epithelial glands. Epithelial cells have small basophilic, irregular nuclei and abundant cytoplasm. Cytoplasm contains muscin. Tumor cells arranged in glands. Mitotic figures are not evident. The tumor cells invaded the muscularis propria and subserosa. The stroma is infiltrated by large numbers of inflammatory cells. Tumor necrosis and blood vessel invasion are not revealed. Lymph nodes show hyperplasia.

  • Purification notesTissue specimens are homogenized in modified RIPA buffer to obtain the soluble proteins, and centrifuged to clarify. The pellet was further extracted with a second buffer to obtain the less soluble protein fraction. The lysate solution may appear turbid at cold temperatures due to insolubility of buffer components. The solution should clear upon warming to room temperature.
