Product overview
The Human Galectin-9 / Gal-9 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213786) is designed for the quantitative detection of Human Galectin-9 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
The ELISA kit is based on standard sandwich enzyme-linked immunosorbent assay technology. A polyclonal antibody from goat specific for Galectin-9 has been precoated onto 96-well plates. Standards (Expression system for standard: E.coli; Immunogen sequence: M1-T355) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Galectin-9 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Galectin-9 amount of sample captured in plate.

Notes
Galectin-9, also called HUAT or LGALS9A is a protein that in humans is encoded by the LGALS9 gene. This gene is mapped to 17q11.2. The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. The protein encoded by this gene is an S-type lectin. It is overexpressed in Hodgkin's disease tissue and might participate in the interaction between the H and RS cells with their surrounding cells and might thus, play a role in the pathogenesis of this disease and/or its associated immunodeficiency. The protein has N- and C- terminal carbohydrate-binding domains connected by a link peptide.

Function
Binds galactosides. Has high affinity for the Forssman pentasaccharide. May play a role in thymocyte-epithelial interactions relevant to the biology of the thymus. Inhibits cell proliferation. It is a ligand for HAVCR2/TIM3. Induces T-helper type 1 lymphocyte (Th1) death. Isoform Short acts as an eosinophil chemoattractant. -
Tissue specificity
Peripheral blood leukocytes and lymphatic tissues. Overexpressed in Hodgkin disease tissue. -
Sequence similarities
Contains 2 galectin domains. -
Domain
Contains two homologous but distinct carbohydrate-binding domains. -
Cellular localization
Cytoplasm. Secreted. May also be secreted by a non-classical secretory pathway. - Information by UniProt
