    ICAM2 in vitro SimpleStep® ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of human ICAM2 protein in serum, plasma, and cell culture supernatants.


    The SimpleStep® ELISA employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material.  TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Intercellular adhesion molecule 2 (ICAM2), also known as CD102, is a ligand for the leukocyte adhesion protein LFA-1 (integrin alpha-L/beta-2). ICAM2 is an integral membrane protein that has 2 immunoglobulin-like domains, and may play a role in lymphocyte recirculation by blocking LFA-1-dependent cell adhesion. It mediates adhesive interactions important for antigen-specific immune response, NK-cell mediated clearance, lymphocyte recirculation, and other cellular interactions important for immune response and surveillance.

    ICAM2 is a member of the intercellular adhesion molecule (ICAM) family. All ICAM proteins are type I transmembrane glycoproteins, contain 2-9 immunoglobulin like C2 type domains, and bind to the leukocyte adhesion LFA1 protein. This protein may play a role in lymphocyte recirculation by blocking LFA1 dependent cell adhesion. It mediates adhesive interactions important for antigen specific immune response, NK cell mediated clearance, lymphocyte recirculation, and other cellular interactions important for immune response and surveillance.

Undiluted samples are as follows: plasma (EDTA) 5%, plasma (citrate) 5%, plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ICAM2 concentration was determined to be 68 ng/mL in plasma (EDTA), 83 ng/mL in plasma (heparin), and 66 ng/mL in plasma (citrate).

  • The concentrations of ICAM2 were measured in duplicates, interpolated from the ICAM2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, and HUVEC supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ICAM2 concentration was determined to be 124 ng/mL in serum, and 9 ng/mL in HUVEC supernatant.

  • Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ICAM2 concentration was determined to be 145 ng/mL with a range of 103-190 ng/mL.
