  • Product overview

Abcam’s RANTES Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human RANTES in serum, plasma, and cell culture supernatants.

This assay employs an antibody specific for Human RANTES coated on a 96-well plate. Standards and samples are pipetted into the wells and RANTES present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human RANTES antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RANTES bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • FunctionChemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. Binds to CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils.
  • Tissue specificityT-cell and macrophage specific.
  • Sequence similaritiesBelongs to the intercrine beta (chemokine CC) family.
  • Post-translational
N-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells.
The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in PubMed:1380064. They are assigned by similarity.

RANTES-CCL5 human macrophage secretion

Excellent Excellent 5/5 (Ease of Use)
RANTES-CCL5 secretion by human macrophages isolated by negative selection in culture for 3 & 7 days.
Kit easy to use, the assay takes about 6hr, good specificity as shown by higher secretion by M1 macrophages (MSCF+IFNg) and M2a (MCSF+IL4) on day 3 compared to the other time point and subtypes (M0=MCSF DEX=MCSF+Dexamethasone IL10=MCSF+IL10 TGFb=MCSF+TGFb)
