  • FunctionSerum albumin, the main protein of plasma, has a good binding capacity for water, Ca(2+), Na(+), K(+), fatty acids, hormones, bilirubin and drugs. Its main function is the regulation of the colloidal osmotic pressure of blood. Major zinc transporter in plasma, typically binds about 80% of all plasma zinc.

  • Involvement in diseaseDefects in ALB are a cause of familial dysalbuminemic hyperthyroxinemia (FDH) [MIM:103600]. FDH is a form of euthyroid hyperthyroxinemia that is due to increased affinity of ALB for T(4). It is the most common cause of inherited euthyroid hyperthyroxinemia in Caucasian population.

Anti-Human Serum Albumin antibody [Alb1] images

  • Cells were grown on glass coverslips, fixed with 4% PFA/PBS for 10 minutes at room temperature and washed once with PBS. They were then treated with 250 mM Tris-HCl pH 8.0 for 10 minutes, washed twice with PBS, and permeabilized with 0.2% TritonX-100 in PBS for 20 minutes at room temperature. Cells were blocked with 5% BSA/0.1%Tween-20/PBS for 30 minutes at room temperature and incubated with ab399 in blocking buffer 1:250 for 1 hour at room temperature, then washed 3 times with PBS. They were then incubated with a goat anti-mouse-Alexa Fluor 488 (1:1000 in Blocking buffer) for 1 hour at room temperature, washed 3 times with PBS, and imaged in PBS. The Huh7.5 cells (human hepatoma, express albumin) showed a pattern of staining consistent with ER, Golgi and secretory vesicle localization. HeLa cells (do not express albumin) did not stain and BSA blocking did not cause nonspecific staining in HeLa cells.

We have no reports of it being used for this application, although with a hign affinity constant we do not see any reason it should not work for this.
