    Abcam’s Triiodothyronine (T3) in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Triiodothyronine (T3) in Human serum.

    A 96-well plate has been pre-coated with Anti-Triiodothyronine antibodies. Samples, standards and the Triiodothyronine HRP Conjugate are added to the wells, where Triiodothyronine in the sample competes with the added Triiodothyronine-HRP for antibody binding. After incubation, the wells are washed to remove unbound material and TMB substrate is then added which is catalyzed by HRP to produce blue coloration. The reaction is terminated by addition of Stop Solution which stops the color development and produces a color change from blue to yellow. The intensity of signal is inversely proportional to the amount of Triiodothyronine in the sample and the intensity is measured at 450 nm.

This ELISA kit was used with cell supernatant and the cell extract of HEK293 after homogenisation. The cells were transfected with Type-II Deiodinase variants and were later treated with a T4 medium at a concentration of 20pM. The kit was successful in measuring the concentration of T3 in both the supernatant and cell extract yielding a range of results from 2.9-8.6pg/mL.

This kit is not sensitive enough to detect any differences in free T3 generated by cells in cell culture supernatant. It can easily detect exogenous T3 added at 20 nM with strong signal.

It seems that mouse serum can be used with this kit, yielding values in the range of 2-6 pg/mL, which is in the range of previously reported values for human free T3 (http://www.globalrph.com/labs_t.htm#T4_(thyroxine)_Total_) and mouse free T3 (http://genesdev.cshlp.org/content/13/10/1329.full - see figure 2A, WT).
