Applications

Our Abpromise guarantee covers the use of ab66653 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Detects a band of approximately 50 kDa (predicted molecular weight: 54 kDa).

ab66653 detects a band at 50 kDa. This band corresponds in size to the mature cleaved form of HYAL2. Abcam recommends using BSA as the blocking agent. Abcam welcomes customer feedback and would appreciate any comments regarding this product.

Target

Anti-HYAL2 antibody images

  • All lanes : Anti-HYAL2 antibody (ab66653) at 1 µg/ml

Lane 1 : HeLa (Human epithelial carcinoma cell line) Whole Cell Lysate
Lane 2 : Thymus (Rat) Tissue Lysate
Lane 3 : Skeletal Muscle (Mouse) Tissue Lysate

Lysates/proteins at 10 µg per lane.

    Secondary
    Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/10000 dilution
    Developed using the ECL technique

    Performed under reducing conditions.

Predicted band size : 54 kDa
Observed band size : 50 kDa (why is the actual band size different from the predicted?)
Additional bands at : 30 kDa,42 kDa,55 kDa (possible immature (unprocessed)). We are unsure as to the identity of these extra bands.

    Exposure time : 4 minutes

This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab66653 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.

Lane 5 : Raji whole cell lysate (ab30124)
    Lane 6 : SW480 whole cell lysate (ab3957)

Lysates/proteins at 10 µg per lane.

    Secondary
Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution

    Performed under reducing conditions.

Predicted band size : 54 kDa
Observed band size : 50 kDa (why is the actual band size different from the predicted?)
Additional bands at : 54 kDa (possible post-translational modification),70 kDa. We are unsure as to the identity of these extra bands.
