Nuclear factor kappa B (NF-kB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kB mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kB is associated with IkB proteins in the cell cytoplasm, which inhibit NF-kB activity. IkB is phosphorylated by IkB kinase(IKK) complex that contains IKKa, IKKb, and IKKg. A novel molecule in the IKK complex was recently identified and designated IKKe/IKK-i. IKKe is required for the activation of NF-kB by PMA and T cell receptors but not by TNFa and IL-1. IKKe/IKK-i message is expressed in a variety of tissues and is inducible by TNFa, IL-1, and LPS. -

Phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. May play a special role in the immune response. Protects cells against DNA damage-induced cell death. -

Highly expressed in spleen followed by thymus, peripheral blood leukocytes, pancreas, placenta. Weakly expressed in lung, kidney, prostate, ovary and colon. -

ab7891 at 10µg/ml staining IKK epsilon in human pancreatic tissue by IHC.

ICC/IF image of ab7891 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7891, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
