Application Abreviews Notes
WB 1/400 - 1/800. Predicted molecular weight: 21 kDa. In Western blot analysis of natural cell products or human body fluids, multiple bands of IL-10 will appear due to the variable amount of glycosylation on the molecule.
IHC-Fr 1/400.
IP 1/400 - 1/800. Pre-clearing with a non-specific rabbit IgG is recommended to reduce background.
ELISA 1/400 - 1/1200. (as detection antibody, using a monoclonal antibody for capture).
Neutralising 1/400. Incubate the samples with the antiserum for at least 4 hours before being tested. A control of similarly diluted normal rabbit IgG (heat inactivated) is recommended.
RIA 1/4000.
IHC-P 1/400.
ICC/IF Use at an assay dependent concentration.

  • ab34843 staining IL10 in Human colon tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).

    Tissue was fixed with paraformaldehyde and blocked with 1% BSA for 20 minutes at 25°C; antigen retrieval was by heat mediation. Samples were incubated with primary antibody (1/200 in 1% BSA in PBS) for 16 hours at 4°C. An undiluted HRP-conjugated goat anti-rabbit polyclonal IgG was used as the secondary antibody.

  • Immunofluorescence analysis of Human peripheral blood mononuclear cells, staining IL10 with ab34843.

    Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 in PBS and blocked with 1% BSA for 1 hour at 25°C. Samples were incubated with primary antibody (1/100 in 1% BSA in PBS) for 24 hours at 4°C. An AlexaFluor®594-conjugated goat anti-rabbit polyclonal IgG (1/250) was used as the secondary antibody.
